چکیده

Consistent performance of anti-drug antibody (ADA) assays through all stages of clinical development is critical for the assessment of immunogenicity and interpretation of PK, PD, safety, and efficacy. The electrochemiluminescent assays commonly employed for ADA measurement use drug conjugated with ruthenium and biotin to bind ADA in samples. Here we report an association between high nonspecific ADA responses in certain drug-naïve individuals and the storage buffer of the conjugated reagents used in a monoclonal antibody ADA assay. Ruthenylated reagents stored in phosphate-buffered saline (PBS) buffer had increased levels of aggregate and produced variable and high baseline responses in some subjects. Reagents stored in a histidine-sucrose buffer (HSB) had lower aggregate levels and produced low sample responses. In contrast to PBS, conjugated reagents formulated in HSB remained low in aggregate content and in sample response variability after 5 freeze/thaw cycles. A reagent monitoring control (RMC) serum was prepared for the real-time evaluation of conjugated reagent quality. Using appropriate buffers for storage of conjugated reagents together with RMCs capable of monitoring of reagent aggregation status can help ensure consistent, long-term performance of ADA methods.

Rheumatoid factor interference in immunogenicity assays for human monoclonal antibody therapeutics.

Rheumatoid factors (RFs) are endogenous human antibodies that bind to human gamma globulins. RFs demonstrate preferential binding to aggregated gamma globulins and are involved in the clearing mechanism of immune complexes. Immunoassays designed to measure human anti-human antibodies (HAHA) after administration of monoclonal antibody therapeutics are thus vulnerable to interference from RFs. Wh...

Synthesis and characterization of physicochemical and immunological properties of recombinant NS3-G2 dendrimer conjugate

Introduction: An effective vaccine against HCV infection is not available. The non-structural protein 3 (NS3) of the virus as an important immunogenic candidate has been utilized in various modules. Nanostructured polymers have been recently used for efficient vaccine and drug delivery. The aim of the current study was the synthesis of rNS3-G2 conjugate and preliminary evaluation of its immunog...

DETERMINATION OF SERUM CORTISOL LEVEL BY A DIREC T ENZYME IMMUNOASSAY USING PENICILLINASE AS LABEL

Serum cortisol level was measured by an enzyme immunoassay (EIA) using the enzyme penicillinase as a label without prior extraction and purification. Polyclonal antibodies were raised against cortisol-3-ortho-carboxymethyl-oxime (cortisol-3-0-CMO) conjugated to bovine serum albumin (BSA). This antibody showed a very low cross-reactivity with structurally related steroids (3.7% for corticos...

کلون‌سازی، بیان و تخلیص پروتئین اینتگراز ویروس HIV و بررسی آنتی‌ژنیسیته آن

Background: Improving the performance of HIV diagnosis assays is one of the most important ways to reduce HIV transmission. Because of the high mutation rate of HIV, it is critical to use the conserved proteins to develop diagnostic immunoassay methods. Because integrase is one of the most conserved proteins of HIV, it may be a good target for this purpose. In this paper cloning, purification a...
